Video Conference between Scientists of StSMU and Germany

November 3, 2015, the StSMU hosted a video conference involving researchers from Germany. Distance learning has become part of the advanced academic technologies in higher education focusing on individual demand expressed by the students, as well as on their major specialty.

IMG_7010.jpgThe Stavropol State Medical University has been successfully carrying out its stage-by-stage implementation of degree programs offering certain elements of remote training employing computer technologies. One of such elements was the video conference and the colloquium-seminar involving Prof. Sergey Sirak and Prof. W.-D. Grimm of the Witten/Herdecke University (North Rhine-Westphalia, Germany) together with clinical interns, resident doctors and dentists representing various clinics of the Stavropol region who undergo training at the Dentistry Department of the StSMU.

This type of interaction stirred great interest in the audience who were eager to learn about the specific features of teaching dentistry in Russia and abroad, as well as to discuss the pros and cons of arranging field placement and practical skills training for students and interns in Russian and German medical schools.

Further on, Prof. Grimm delivered to the doctors, interns, and residents an interactive colloquium-seminar, which is part of the classes aiming at deeper understanding and mastering of innovation technologies in treating affected tissues of periodontium and using cell technologies in dental implantology.

evaluated in vitro and ex vivo

13-18. 05. 2015

Objectives:

The aim of the Secound International Symposion “Age of Regenerative Medicine "

organized and provided by the Stavropol State Medical University 2015 will be to prepare methods and aims for investigating the proliferation and differentiation of a human ecto-mesenchymal Stem Cells cell lines derived from Human Palate in vitro and ex vivo.

Background data:

Regenerative medicine is currently focused on directing differentiation of stem cells into lineage-specific, functional cells that can promote tissue repair and organization. Stem cell therapies are attractive for treating numerous diseases, including dental diseases (for example, tooth decay, periodontal diseaeses, and for improvement of preimplantological bone situations), which are among the most prevalent health concerns. Current clinical dentistry is predominantly focused on restorative approaches involving placement of inert materials, but tissue regeneration is an attractive alternative because materials alone fail with time and do not provide the full function of the tissue.

First Topic of the Symposion: Proliferation and Differentiation of human ecto-mesenchymal Stem Cells from Human Palate evaluated in vitro

The autonomous migration of specialized cells is an essential characteristic in both physiological and pathological functions in the adult human organism. Leukocytes, fibroblasts, and stem cells, but also tumor cells, are thus the subject of intense investigation in a broad range of research fields. A wide spectrum of methods have therefore been established to analyze chemokinetic and chemotactic cell migration, ranging from easy-to-handle two-dimensional surface migration assays to highly specialized three-dimensional and intravital analysis methods. It is now manifest that the results obtained with these various migration assays substantially differ.

Targeting resident stem cell pools by activating endogenous morphogens could potentially bypass many of the limitations of the current approaches to regenerative medicine and revolutionize the field. Among these endogenous factors, physiologically available latent growth factor complexes, such as members of the transforming growth factor–ß (TGF- ß) superfamily, could be harnessed as potent cues. TGF- ß is secreted as a latent complex, and its activation is a key step in its physiological function. The TGF- ß superfamily is encoded by 23 distinct genes that include TGF- ß, bone morphogenetic protein (BMP), Activin, Nodal, Inhibin, and growth differentiation factor (GDF). Their interactions and complex context-dependent roles in modulating stem cell pluripotency and differentiation have been elegantly demonstrated in a variety of experimental models (Widera et al. 2007, Grimm et al. 2011, Keeve et al. 2013). Recent studies highlight the role of the TGF-ß signaling pathway as a master regulator of stem cell differentiation (see overview Widera et al. 2004). The induction of Oct4 and Nanog expression by specific inhibition of TGF- ß receptor I (TGF- ßRI) with subsequent absence of lineage differentiation marker expression has identified TGF- ß signaling as a key player in stem cell pluripotency and differentiation (see overview Widera et al. 2008). Moreover, TGF-ßs have a central role in the periodontal and periimplant pathophysiology that is used as the experimental model in this study, and are among the most promising cues in regenerative periodontology and dental implantology.

Second Topic of the Symposion: Isolation and Differentiation of Palatal-derived ecto-mesenchymal Stem Cells for Regenerative Procedures in Dentistry

Several different biological and biomechanical cues are presently being explored to direct stem cell behavior. Due to their broad differentiation potential and their persistence into adulthood, human neural crest-derived stem cells (NCSCs) harbour great potential for autologous cellular therapies. Clinical application of stem cell therapies is exemplified by culture-based manipulation of patient-specific cells and reintroduction into specific anatomical contexts. In contrast, exogenous morphogens may be delivered to tissues to mobilize and promote resident stem cells toward specific biological functions. In either situation, exogenous materials must be delivered to the body with stringent spatial and temporal control, and this poses significant technical, regulatory, economic, and biological (off-target effects) barriers to widespread clinical applications.

Third Topic of the Symposion: Translational Research: Palatal-derived ecto-mesenchymal Stem Cells from Human Palate: A New Hope for Alveolar Bone Regeneration

The management of facial defects has rapidly changed in the last decade. Functional and esthetic requirements have steadily increased along with the refinements of surgery. In the case of advanced atrophy or jaw defects, extensive horizontal and vertical bone augmentation is often unavoidable to enable patients to be fitted with implants. Loss of vertical alveolar bone height is the most common cause for a non primary stability of dental implants in adults. At present, there is no ideal therapeutic approach to cure loss of vertical alveolar bone height and achieve optimal pre-implantological bone regeneration before dental implant placement. Using our research data we have started a proof of principle clinical- and histological controlled study using stem cell-rich palatal tissues for improving the vertical and the lateral alveolar bone augmentation in critical size alveolar defects.
